摘要
目的:为提高白花蛇舌草(HD)总黄酮的纯度,筛选适宜HD总黄酮纯化的大孔树脂,确定HD总黄酮纯化工艺参数。方法:采用静态与动态吸附-解吸附方法对HD总黄酮进行纯化,以吸附率与洗脱率等指标优化工艺,利用紫外-可见分光光度计测量HD总黄酮的含量。结果:HPD-826型大孔树脂纯化效果最好,其最佳工艺为上样流速2 BV.h-1,上样液pH 5,上样浓度1.5 g.L-1,洗脱剂pH 7,乙醇浓度80%与洗脱速度2 BV.h-1;纯化后总黄酮含量高达91.24%。结论:该工艺操作简单、稳定可行,重复性好,适合HD总黄酮纯化。
Objective:Purify the total flavonoids from Hedyotis diffusa, with macroporous resin and establish optimum process for separating and purifying total flavonoids from H. diffusa. Method: The optimal techniques of separation and purification of flavonoid were obtained by static adsorption and elution tests, and the separation techniques were evaluated by measuring the concentration of total flavonoids from H. diffusa with ultraviolet spectrophotometer. Result: The HPD-826 macroporous resin gave the best effect of separation with the sampling rate 2 BV, h ^-1 , concentration 1.0 g· L^-1 , pH 5 respectively, and the concentration of elution was 80% alcohol, eluting velocity 2 BV·h^-1, with pH 7. Conclusion: HPD-826 resin could be applied to purify total fiavonoids from H. diffusa.
