摘要
采用荧光定量实时RT-PCR方法,137Csγ射线全身慢性照射SD大鼠,研究其血液中白细胞CDKN1A基因mRNA表达水平的变化;测定了转入慢性期的事故受照人员外周血白细胞CDKN1A基因表达水平。结果表明:与对照组相比,0.1、0.2和3.0 Gy照射组大鼠CDKN1A基因mRNA表达水平降低,与事故急性受照人员转为慢性期后的结果一致;照射组大鼠白细胞数有降低的趋势,淋巴细胞百分比降低,中性粒和单核细胞百分比增加。结果提示,白细胞计数降低及分类发生变化可能是CDKN1A表达水平降低的原因之一,慢性照射和急性照射的基因表达模式可能不同。
The aim of the present study was to investigate the expression of CDKNIA gene in leukocytes of rats irradiated chronically with different doses of gamma rays and in leukocytes of patients accidentally exposed to 60Co gamma rays. Real time SYBR Green I quantitative RT-PCR was adopted to quantify mRNA levels. The results showed that the mRNA levels of CDKN1A gene lowered in 0.1, 0.2 and 3 Gy irradiated groups of rats. The finding was consistent with the results in peripheral leucocytes of patients at the time of half and one year after accidental exposure. It is different from that of increase of CDKN1A expression in acute radiation according to literature. Furthermore, there was a decreasing trend in leucocytes in chronically-irradiated rats, different from the ephemeral increase in acute radiation. The decrease of leucocytes and changes in percentage of leukocyte types would contribute to the decreasing of CDKN1A expression. The findings suggest that the difference in gene expression profile exists between acute radiation and chronic radiation.
