牛布氏杆菌间接ELISA检测方法的建立 被引量:13

Development of indirect ELISA for detection of Brucella abortus

摘要 采用原核表达并纯化的布氏杆菌周质蛋白BP26作为包被抗原,建立了检测牛布氏杆菌血清抗体的间接ELISA(iELISA)方法。用建立的iELISA方法对138份临床奶牛血清样本进行了检测,并与虎红平板凝集试验(RBPT)和套式PCR(nested-PCR)进行了比较。结果表明,所建立的iELISA与RBPT和nes-ted-PCR的符合率分别为97.82%和98.55%。尽管该方法的敏感性略低于nested-PCR,但用iELISA检测病牛血清的阳性率可达93.00%。该iELISA方法可作为一种有效的牛布氏杆菌病血清学诊断的备用方法。 An indirect enzyme-linked immunosorbent assay(iELISA) for detecting antibodies to Brucella abortus was established using the purified recombinant protein BP26 of B.obortus expressed in Escherichia coli as coating antigen.Serum samples(n=138) from dairy cow were simultaneously tested by the iELISA,the rose Bengal plate test(RBPT) and the nested-PCR.The coincidence of iELISA with RBPT and nested-PCR was 97.82% and 98.55%,respectively.Although this iELISA method appeared less sensitive than nested-PCR with positive rate of 93% in dairy cows infected by B.abortus.The iELISA could be used as an alternative method for serological diagnosis of brucellosis.
