摘要
目的:探索应用亚硝基胍(NTG)诱导鼠伤寒沙门菌(ATCC14028)形成营养缺陷型细菌,以观察其对人结肠癌细胞株SW480裸鼠模型的治疗作用。方法:用NTG诱变鼠伤寒沙门菌(ATCC14028)后,用葡萄糖基础培养基+氨苄青霉素筛选出营养缺陷型沙门菌,将其静脉注射入10只无瘤裸鼠尾静脉,观察其毒性;同时将其注入20只荷瘤裸鼠中观察其治疗作用。结果:1只注射入营养缺陷型沙门菌的裸鼠死亡,而注射野生型沙门菌的裸鼠均死亡。20只荷瘤鼠1周后肿瘤组织均出现大小不等的坏死灶。结论:营养缺陷型沙门菌对裸鼠毒性弱,对肿瘤组织有趋化性及肿瘤杀灭作用。
Objective Exploring the treatment to tumor-bearing mice of SW480 human colon cancer cell with auxotrophy Salmonella typhimurium which was mutagenized by N-methyl-N'-nitro-N-nitrosoguanidine(NTG).Methods after using NTG mutagenize Salmonella typhimurium,the auxotrophs was isolated through glucose minimal broths culture and ampicillion selection.It was injected into the tail vein of 10 no tumor-bearing nude mice to see its safty,and injected into 20 tumor-bearing nude mice to observe its anti-tumor effect.Results Contrast to all death of no tumor-bearing nude mice injected with wild type,only one mouse by the auxotrophs died.One week later,all 20 tumor-bearing mice have necrotic areas in tumor after injected by the auxotrophs.Conclusions The auxotrophs is safe for normal mice,and have chemotaxis and anti-tumor effect toward cancer foci.
