摘要
目的:探讨控制性促排卵(contral ovarian stimulation,COS)黄体期添加雌激素对小鼠子宫内膜白血病抑制因子(leukemia inhibitory factor,LIF)mRNA表达的影响。方法:将正常雌性昆明小鼠随机分为A组(单纯COS组)、B组(COS+黄体酮组)、C组(COS+黄体酮+雌激素组)和D组(自然周期,对照组),采用逆转录聚合酶链反应法检测LIF mRNA在各组小鼠妊娠第3~5天子宫内膜的表达情况。结果:4组小鼠妊娠不同时间子宫内膜LIF mRNA表达差异均有统计学意义(P〈0.05);B,C,D组妊娠第4天时LIF mRNA表达水平均达峰值,而A组妊娠第3天其表达水平最高;妊娠小鼠子宫内膜种植窗高峰日LIF mRNA的表达,C,D组均明显高于B组(P〈0.05),B组高于A组(P〈0.05)。结论:添加雌激素较传统黄体支持更能改善小鼠子宫内膜LIF mRNA在小鼠子宫内膜的表达,增加子宫内膜容受性,纠正控制性促排卵对内膜种植窗前移的负面影响。
Objective To evaluate the effect of estradiol on the expression of leukemia inhibitory factor mRNA in the endometrium of mice during the luteal phase of controlled ovarian stimulation cycles.Methods Female mice were randomly divided into group A(contral ovarian stimulation group),group B(contral ovarian stimulation with progesterone for luteal-phase-support),group C(contral ovarian stimulation with progesterone and estradiol for luteal-phase-support) and group D(nature cycle without drugs).The expression of leukemia inhibitory factor mRNA was determined by reverse transcriptional-polymerase chain reaction in the uterine endometrial of pregnant mice on the third to fifth pregnant days.Results There were significant differences in the expressions of leukemia inhibitory factor mRNA in endometrium in different pregnant days in different groups(P0.05).The peak of leukemia inhibitory factor mRNA expression in group B,C and D was on the forth day,and was on the third day in group A.In group C and D,the expressions of leukemia inhibitory factor mRNA in endometrium were significantly higher than those in group B(P0.05),and were higher in group B than those in group A(P0.05).Conclusion Estradiol supplementation during the luteal phase can improve the expression of leukemia inhibitory factor mRNA in the endometrium more effectively than progesterone supporting only,and then improve the endometrial receptivity.
