人翼状胬肉中Rac1蛋白和基质金属蛋白酶-2的表达及临床意义 被引量:1

摘要 目的研究Rac1蛋白和基质金属蛋白酶-2(MMP-2)在人翼状胬肉中的表达,分析二者的相关性,探讨利用Rac1及MMP-2作为标记物评估翼状胬肉生物学特性,为临床诊疗提供参考。方法应用辣根过氧化物酶标记的链霉卵白素免疫组织化学方法(SP法)对39例原发性翼状胬肉患者球结膜组织及12例正常球结膜组织中的Rac1蛋白及MMP-2蛋白进行检测。结果 Rac1蛋白及MMP-2蛋白在翼状胬肉组织中的表达均高于正常结膜组织,差异有显著性意义(Z=-3.386、-3.044,均为P<0.05)。翼状胬肉患者Rac1及MMP-2蛋白表达呈显著正相关(r=0.571,P<0.05)。结论 Rac1及MMP-2在翼状胬肉中的表达,提示翼状胬肉在细胞骨架,基底膜分解代谢和细胞增殖活跃方面发生了变化。Rac1/MMP-2通路在翼状胬肉的发生、发展中起着重要作用。 Objective To investigate expression of Rac1 and matrix metalloproteinase-2(MMP-2)in human pterygium.The relationship among them was analyzed.The possibility was discussed for utilizing Rac1 and MMP-2 as markers for assessment of biological characteristics of pterygium to provide reference for clinical practice.Methods Horseradish peroxidase labeled strepto-antibiotin immunohistochemistry was used to measure the expression of Rac1 and MMP-2 in bulbar conjunctiva of 39 cases with primary pterygium and 12 normal cases.Results The expression of Rac-1 and MMP-2 in primary pterygium were higher than normal conjunctiva,and the difference was significant(Z=-3.386,Z=-3.044;both P0.05).Expression of Rac1 and MMP-2 was positively correlated(r=0.571,P0.05).Conclusions Expression of Rac1 and MMP-2 in pterygium suggests that there are some changes in cell cytoskeleton,decomposition metabolism of basement membrane and cell proliferation in pterygium.Rac1/MMP-2 signaling pathway plays a positive role in pterygium occurrence and development.
