摘要
目的探讨阿魏酸钠(sodium ferulate,SF)对实验性高眼压视网膜缺血-再灌注(retinal ischemic reperfusion,RIR)损伤的保护作用及机制。方法采用眼内灌注法,前房灌注生理盐水建立RIR损伤模型。32只新西兰大白兔随机分为SF治疗组、生理盐水对照组,每组16兔。2组于再灌注后立即按50mg.kg-1体质量耳缘静脉推注25g.L-1SF和生理盐水,每隔24h推注相同量的药物和生理盐水。每组分别于推注后24h、72h空气栓塞法处死动物,用硫代巴比妥分光光度法(TBA)测定丙二醛(malondialdehyde,MDA)含量,黄嘌呤氧化酶法测定超氧化物歧化酶(superoxide dismutase,SOD)活性。结果再灌注24h后,对照组视网膜MDA含量为(3.434±1.003)μmol.g-1,较治疗组(1.394±0.564)μmol.g-1升高(P<0.05),SOD含量为(70.734±1.329)U.mg-1,低于治疗组(98.892±8.979)U.mg-1(P<0.05);再灌注72h后,对照组视网膜MDA含量为(2.639±1.412)μmol.g-1,高于治疗组(1.459±1.552)μmol.g-1(P<0.05),SOD含量为(113.169±0.072)U.mg-1,低于治疗组(139.222±10.066)U.mg-1(P<0.05)。结论 SF注射液可通过降低视网膜组织MDA含量,提高SOD活性,对新西兰大白兔实验性高眼压RIR损伤起一定的保护作用。
Objective To investigate the protective effect and mechanism of sodium ferulate(SF)on retinal ischemia-reperfusion(RIR)injury induced by high intraocular pressure(IOP).Methods The experimental models of RIR injury were established by perfusing physiological saline into the anterior chamber.Thirty-two rabbits were randomly divided into SF treatment group and physiological saline control group,16 rabbits in each group.25 g·L-1 SF was intravenously and immediately injected with the dosage of 50 mg·kg-1 after reperfusion in SF treatment group,and the same capacity of physiological saline was given in control group,the same capacity was given per 24 hours in two groups.The rabbits were executed by aeroembolism at 24 hours,72 hours after injection.The content of malondialdehyde(MDA)and the superoxide dismutase(SOD)activity were measured by TBA and xanthine oxidase methods,respectively.Results At 24 hours after reperfusion,the content of MDA in control group and treatment group were(3.434±1.003)μmol·L-1 and(1.394±0.564)μmol·L-1,the later was lower than the former(P0.05);The activity of SOD in control group and treatment group were(70.734±1.329)U·mg-1 and(98.892±8.979)U·mg-1,the latter was higher than the former(P0.05).At 72 hours after reperfusion,the content of MDA in control group and treatment group were(2.639±1.412)μmol·L-1 and(1.459±1.552)μmol·L-1,the latter was lower than the former(P0.05);The activity of SOD in control group and treatment group were(113.169±0.072)U·mg-1 and(139.222±10.066)U·mg-1,the latter was higher than the former(P0.05).Conclusion SF has the protective effect on experimental RIR injury induced by high intraocular pressure by increasing the activity of SOD and decreasing the content of MDA.
