摘要
目的:观察结直肠癌LoVo细胞株发生上皮细胞间质转型(EMT)逆转前后3种耐药相关ABC转运蛋白基因的表达.方法:运用荧光定量PCR的方法,比较发生EMT的细胞株LoVo GnT-V/NC和EMT逆转之后的细胞株LoVo GnT-V/1564中 ABCB1、ABCC3、ABCB6基因表达量.结果:LoVo GnT-V/NC细胞中ABCB1基因表达量是LoVo GnT-V/1564的7.67倍;LoVo GnT-V/NC和LoVo GnT-V/1564细胞中ABCC3、ABCB6基因表达量差异均无统计学意义(P〉0.05).结论:LoVo细胞株EMT发生后肿瘤细胞的耐药相关基因ABCB1的表达量也发生了改变,EMT可能与肿瘤的多药耐药相关.
Objective:To study the expression of ABC transport protein gene in colorectal cancer LoVo cell line before and after epithelial-mesenchymal transitions (EMT). Methods: FQ-PCR (fluorescence quantitative polymerase chain reaction) was used to compare the expression of ABCBI ,ABCC3 and ABCB6 genes in LoVo GnT- V/NC cell line with EMT and in LoVo GnT-V/1564 cell line after EMT inversion. Results: The expression of ABCB1 in LoVo GnT-V/NC cells was 7.67 times to LoVo GnT-V/1564, while no statistical difference was found in the expression of ABCC3 and ABCB6 between LoVo GnT-V/NC and LoVo GnT-V/1564 (P 〉 O. 05). Conclusions : Expression of drug resistance related gene ABCB1 changed after EMT of LoVo cell line, indicating that EMT may be associated with multi-drug resistance (MDR) of tumor.
