5-氮杂胞苷和藤黄酸对K562/A02耐药细胞株的抑制作用 被引量:4

Inhibitory effect of 5-AZA and gambogic acid on apoptosis of K562/A02 line

摘要 目的探讨5-氮杂胞苷(5-AZA)协同藤黄酸对K562/A02耐药细胞株的抑制作用。方法采用MTT比色法测定柔红霉素(DNR)的半数抑制量(IC50);流式细胞术(FCM)检测细胞凋亡;RT-PCR法检测mdr-1基因mRNA表达。结果 DNR对K562/A02的IC50为19.4 mg/L。5-AZA、藤黄酸和两药联合处理K562/A02细胞后,DNR的IC50值分别为11.1 mg/L、10.5 mg/L和7.1 mg/L。5-AZA与藤黄酸联合应用可增加K562/A02的凋亡。分别单独应用5-AZA、藤黄酸作用后耐药株mdr-1 mRNA下调微弱,两药联合作用后耐药株mdr-1 mRNA下调。结论 5-AZA、藤黄酸均能抑制耐药白血病细胞株K562/A02细胞,联合使用具有协同作用。 Objective To investigate the inhibitory effect of 5-AZA and gambogic acid(GA) on multidrug resistance cell line K562/A02.Methods The IC50 of daunorubicin was assayed by MTT method.The rate of apoptosis of K562/A02 was detected by flow cytometry.The expression of mdr-1 mRNA was assayed by RT-PCR.Results IC50 of DNR on K562 /A02 cells was 19.4mg/L,which was 11.1mg/L for 5-AZA,10.5mg/L for GA and 7.1mg/L for combined use of 5-AZA with GA.The rate of apoptosis was increased by combined use of 5-AZA with GA.Conclusion Multi-drug resistance cell line K562 /A02 can be effectively inhibited by 5-AZA and GA.The combination of both drugs shows a great synergistic reversal effect.
