BTLA mRNA在实验变态反应性神经炎中的动态表达

Dynamic expression of B and T lymphocyte attenuator mRNA in experimental allergic neuritis

摘要 为观察BTLA mRNA在EAN大鼠坐骨神经、脾脏、外周血及淋巴结中的动态表达,探讨BTLA与EAN的发病关系,以提供治疗自身免疫性疾病新的靶点。采用72只Lewis大鼠,SPF级,雄性,6-8周,体重160~180 g,随机分为生理盐水对照组(NS组)、完全弗氏佐剂组(CFA组)及抗原注射组(EAN组)。观察免疫后大鼠的发病情况和组织病理学改变。分别在第7天、第16天、第24天、第33天处死动物,取坐骨神经根制作病理切片行HE染色和Weil染色观察其病理改变,利用RT-PCR检测坐骨神经、脾脏、外周血和淋巴结中BTLA mRNA的表达水平。结果是EAN组大鼠在发病16d症状达高峰,第33天基本好转。与对照组相比,EAN组大鼠各组织各时间点BTLA mRNA表达均增高(P〈0.05),整个实验过程中一直呈上升趋势;CFA组和NS组大鼠无症状,CFA组与NS组相比,BTLA的表达无明显差异。各组织中BTLA的表达具有相关性。提示BTLA在EAN发病过程中的动态变化可能参与EAN的发病。 The study on the involvement of B and T lymphocyte attenuator(BTLA) in experimental allergic neuritis(EAN) will give us new target for the treatment of autoimmune disease.In the present study,72 male Lewis rats were selected and divided into 3 randomized groups,i.e.NS group,CFA group,and EAN group.The rats in the immunized and control group were sacrificed on the 7 day,16 day,24 day,and 33 day post immunization(p.i).Pathologic paraffin sections were taken from sciatic nerve roots of rats and stained with HE and Weil's stain.Reverse transcriptase polymerase chain reaction(RT-PCR) was used to detect the level of BTLA mRNA in spleen,sciatic nerve,peripheral blood lymphocytes and lymph nodes.It was found that the clinical manifestations attained the peak of clinical score on the 16 day p.i,and ameliorated obviously on the 33 day p.i.In comparison with the control group,the level of BTLA mRNA was up-regulated in the whole process of experiment(P0.05).No clinical manifestation was demonstrated in CFA and control group of rats.Nevertheless,correlation in BTLA expression existed in various tissues of rats,From these observations,it is clear that the dynamic expression of BTLA mRNA in the course of disease may play a role in the pathogenesis of EAN.
