摘要
本文以市售海带为原料,通过单因素试验选取了试验的因素与水平,在单因素试验基础上,通过正交试验确定了褐藻酸提取的最佳工艺条件。并采用酸法降解褐藻酸得到降解多糖D-AGS。D-AGS经过DEAE-Sephadex A-25凝胶柱和Sephadex G-75凝胶柱分离纯化后,得到两个单一组分D-AGS-1A和D-AGS-2A,并对D-AGS-2A进行了理化性质研究。通过紫外光谱表明其不含有核酸和蛋白质,碘与碘化钾反应、斐林反应、塞式、考马斯亮蓝反应均呈阴性,咔唑-硫酸反应呈阳性。
Taking Laminaria japonica as raw material,extraction of alginic acid was studied and optimized by L9(34) orthogonal test.And D-AGS was obtained by the degradation of alginic acid,which was further purified by chromatography on DEAE-Sephadex A-25 and Sephadex G-75 columns,giving two pure polysaccharides(D-AGS-1A and D-AGS-2A).The result of UV spectrophotometric illustrated that D-AGS-2A does not contain protein and nucleic acid.And D-AGS-2A was assayed by means of iodine-potassium iodide,Fehling,Seliwanoff,coomassie brilliant blue G-250 and carbazole sulfuric acid reaction.
