摘要
骨性关节炎(osteoarthrits OA)是以关节软骨缺损为特点的一种退行性的骨关节疾病,其病因目前尚不十分清楚,但多种蛋白水解酶活性增高而引起的软骨外基质(extracellular matrix,ECM)的降解异常是软骨退变的重要原因,其中软骨蛋白聚糖酶(aggrecanase)引起的软骨蛋白聚糖降解在骨性关节炎早期阶段是一个重要的事件,此酶(特别是ADAMTS-4和ADAMTS-5)在骨性关节炎软骨中对软骨蛋白聚糖的降解起到重要作用,已经得到大家广泛的关注,目前已将它作为治疗OA的新靶点。此综述将此类酶与OA的关系及在OA中的作用及研究进展作一归纳。
Osteoarthritis (OA) is a degenerative disease of the joints characterized by breakdown of articular cartilage. The pathogenesis of OA is poorly understood, bui the abnormal degradation of extracellular matrix ( ECM), which caused by increased activities of various proteinases, is the important cause in degradation of cartilage. Aggrecanase-mediated aggrecan degradation is the key molecular event in early-stage of OA, and especially the aggrecanases ADAMTS-4 and ADAMTS-5, play a significant role in degradation of aggrecan. There have been considerable interests in the possible role of these aggrecanases as therapeutic targets in OA. This review discusses some recent data regarding aggrecanases in OA, with emphasis on the roles and relationship between aggrecanase and aggrecan in OA.
