摘要 目的观察galectin-1表达对LoVo细胞表型的影响。方法构建galectin-1真核表达载体,转染LoVo细胞,细胞免疫化学检测蛋白质水平的改变,dot-blotting检测细胞培养上清分泌性galectin-1,观察LoVo细胞表型变化。结果成功构建了galectin-1真核表达载体,并建立了稳定的真核表达载体转染LoVo细胞系。galectin-1表达可降低LoVo细胞与Ⅰ型胶原的粘附,促进细胞同质粘附,且降低细胞体内成瘤(P<0.001)。结论Galectin-1表达可降低LoVo细胞恶性相关表型。 Objective To observe the effect of galectin-1 expression on the phenotypes of LoVo cell. Methods Eukaryotic expression vector of galectin-1 was constructed and transfected into LoVo cell using LipofectamineRM 2000.Immunochemistry and dot-ELISA were used to detect the intracellular and extracellular galectin-1.Phenotypes of LoVo cell including heterotypic cell adhesion, homotypic cell adhesion and tumorigenicity assay were observed. Results Eukaryotic expression vector of galectin-1 was successfully constructed.Galectin-1 expression inhibited LoVo cell heterotypic adhesion,accelerated its homotypic adhesion,and attenuated tumorigenicity of LoVo cell(P0.001). Conclusion Galectin-1 expression could decrease LoVo cell malignant phenotypes.
