络气郁滞/虚滞型动脉粥样硬化模型NEI网络指标与血管内皮功能的关系 被引量:5

Correlation between Neuro-Endocrine-Immune Network Indices and Vascular Endothelial Functions in the Atherosclerosis Model with Collateral-qi Stagnation/Deficient Stagnation Syndrome Type

摘要 目的从神经内分泌免疫网络(NEI)角度研究络气郁滞/虚滞证候因素对动脉粥样硬化(AS)的影响。方法建立络气郁滞/虚滞证候模型、大鼠AS病理模型及络气郁滞/虚滞型AS复合模型,并给予通心络胶囊及辛伐他汀进行药物干预,分别检测NEI网络指标及内皮功能障碍指标,分析各模型大鼠NEI网络指标与内皮功能指标之间的相关关系。结果 NEI网络相关因子血管紧张素(Ang)、醛固酮(ALD)、甲状腺素(T4)与一氧化氮(NO)呈正相关,促甲状腺激素(TSH)、白介素-1β(IL-1β)、白介素-2(IL-2)、三碘甲状腺原氨酸(T3)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)、肿瘤坏死因子-α(TNF-α)、促肾上腺皮质激素释放激素(CRH)、血浆肾素活性(PRA)、白介素-6(IL-6)与NO呈负相关。TNF-α、IL-6、PRA、CORT、IL-1β、IL-2、ACTH、CRH、T3、T4与内皮素(ET)呈正相关,ALD、Ang、TSH与ET呈负相关。各药物干预组内皮功能和NEI网络指标模糊聚类结果显示,通心络大剂量组与正常组相似度最高。结论络气郁滞与络气虚滞证候因素均可通过影响NEI网络稳态失衡加重血管病变,通心络胶囊显示出不同于西药的干预NEI网络稳态失衡的独特作用。 Objective To study the influence of collateral-qi stagnation /deficient stagnation syndrome type on atherosclerosis(AS)from the angle of neuro-endocrine-immune(NEI)network.Methods Collateral-qi stagnation /deficient stagnation syndromes model,AS model,and collateral-qi stagnation /deficient stagnation compound with AS model were established with Wistar rats.They were administered Tongxinluo Capsule(Capsule for treating cardiovascular diseases)and Simvastatin for intervention.Their NEI network indices and endothelial dysfunction indices were detected respectively to analyze the correlations between the NEI network indices and endothelial dysfunction indices of all rat models.Results The related factors of NEI network including angiotensin Ⅱ(Ang Ⅱ),aldosterone(ALD),and thyroxine(T4)had positive correlation with nitric oxide(NO),while thyroid-stimulating hormone(TSH),interleukin-1β(IL-1β),interleukin-2(IL-2),triiodothyronine(T3),adrenocorticotrophic hormone(ACTH),cortisone(CORT),tumor necrosis factor-α(TNF-α),corticotropin releasing hormone(CRH),plasma renin activity(PRA),and interleukin-6(IL-6)had a negative correlation with nitric oxide(NO).TNF-α,IL-6,PRA,CORT,IL-1β,IL-2,ACTH,CRH,T3,and T4 had positive correlation with ET,while ALD,Ang Ⅱ,and TSH had a negative correlation with ET.The result of fuzzy clustering of endothelial function and NEI network indices in each intervention group showed that the Tongxinluo large-dose group was in the highest similarity with the normal group.Conclusion The syndrome factors of collateral-qi stagnation /deficient stagnation can make worse vascular lesions through affecting the NEI network steady-state.Different from western medicine,Tongxinluo Capsule has specific intervention on the imbalanced NEI network steady-state.
