摘要
目的:建立同时测定泽泻药材中泽泻醇A、泽泻醇F、24-乙酰泽泻醇A和23-乙酰泽泻醇B含量的HPLC方法。方法:采用RP-C18色谱柱(6.0 mm×150 mm,5μm),乙腈-水梯度洗脱,检测波长210 nm,正交实验优化药材提取方法并测定18个不同批次样品的含量。结果:泽泻醇A、泽泻醇F、24-乙酰泽泻醇A和23-乙酰泽泻醇B在测定的范围内均表现出良好的线性(r〈0.999);方法的回收率在96.67%99.61%,RSD均小于3.0%。结论:该分析方法能简便快速的测定泽泻中多个主要成分的含量,可以较全面的反映泽泻药材质量的优劣。
Objective: To develop an HPLC method for the determination of alisol A,alisol F,alisol A 24-acetate and alisol B 23-acetate in the rhizome of Alisma orientalis.Method: The separation was performed on a Shim-Pack CLC-ODS a C18 column(6 mm×150 mm,5 μm) eluted with the mobile phases of acetonitrile(A)-water(B) in gradient elution.The absorbance was monitored at 210 nm.Orthogonal test was adopted to optimize the extraction conditions of the four alisols.Result: The correlation coefficients of the four alisols were higher than 0.999 and the average recoveries were 99.23%,96.67%,97.30% and 99.61%,respectively.All the RSDs were less than 3%.Conclusion: The validation data demonstrated that the method was accurate and repeatable,and can be use to measure the four alisols in Rhizoma Alismatis.
