HPLC法测定国产尼美舒利血浆浓度

Determination of Nimesulide in plasma by HPLC

目的 :建立了人血浆中尼美舒利浓度的HPLC测定法。方法 :以对羟基联苯为内标 ,甲醇 -水 -乙酸 ( 60∶4 0∶1 )为流动相 ,紫外检测波长为 2 3 0nm。结果 :实验表明 ,尼美舒利与内标分离良好 ,血浆中尼美舒利平均回收率97% ,日内、日间RSD小于 1 2 % ,血浆尼美舒利浓度在 0 .1～ 2 0 μg/ml范围内线性关系良好 ,r=0 .9998。本法的最低检测浓度为 2 0ng/ml。结论 :该法操作简便、结果可靠 。

OBJECTIVE:A rapid,simple and sensitive reversed-phase HPLC method was established for the determination of nimesulide in human plasma.METHOD:The mobile phase consisted of methanol-water-acetic acid(60∶40∶1).The detection wave length was 230nm and flow rate was 1ml/min.Internal standard was p-hydroxy-biphenyl.RESULTS:The retention time of nimesulide and p-hydroxy-biphenyl was 6.07 and 8.10min,respectively.The mean recovery of nimesulide was 102%.The within day and day-to-day of variations were all less than 10%.The detection limit of nimesulide was 20ng/ml in plasma.Nimesulide and propafenone were seperated well.The assay linearity was determined over the range of 0.1～20μg/ml in plasma(r=0.9997).CONCLUSION:The method is sensitive with constant result.It is good for the determination of nimesulide concentration.
