针刀松解法对第三腰椎横突综合征模型兔血清SOD、MDA和T-AOC的影响 被引量:9

Influences of acupotomy therapy on serum SOD,MDA and T-AOC in rabbits with the third lumbar transverse process syndrome

摘要 目的观察第三腰椎横突综合征模型兔血清超氧化物歧化酶(SOD)、丙二醛(MDA)和总抗氧化能力(T-AOC)的变化,探讨针刀松解法干预的作用机制。方法将30只日本大耳白兔随机分5组:正常组、模型组、针刀组、电针组、针刀加电针组,每组6只。后4组采用在左侧第三腰椎横突部埋置明胶海绵的方法制作第三腰椎横突综合征模型,针刀组和电针组在造模后第15天分别给予针刀松解法和电针双侧"委中"干预,针刀加电针组则同时给予针刀松解法和电针干预。造模后第28天,腹主动脉取血,检测血清SOD、MDA和T-AOC的变化,HE染色观察局部组织的病理学改变。结果模型组兔血清中SOD、T-AOC活性较正常显著降低,MDA含量较正常显著升高,有统计学差异(P<0.01,P<0.05),针刀组、电针组、针刀加电针组SOD、T-AOC、MDA与正常组之间均无显著性差异(P>0.05)。针刀组SOD、T-AOC活性较模型组显著升高,MDA含量较模型组显著降低(P<0.05)。电针组、针刀加电针组SOD活性较模型组显著升高(P<0.05,P<0.01),针刀加电针组MDA含量较模型组显著降低(P<0.05)。HE染色显示模型组局部出现明显炎症反应3,个治疗组局部炎症反应均有减轻。结论针刀松解法或(和)电针干预可通过减轻局部炎症反应和提高模型兔血清中SOD和T-AOC活性、降低血清中的MDA含量,有效地清除体内过多的氧自由基,防止组织的进一步损伤,促进损伤组织的修复;且针刀松解法的作用优于电针干预。 Objective To observe the changes of serum superoxide dismutase(SOD),malondialdehyde(MDA) and total antioxidant capacity(T-AOC) in rabbit model of the third lumbar transverse process syndrome,and explore the mechanism of the intervention of acupotomy therapy.Methods Japanese rabbits(n=30) were randomly divided into the normal group,model group,acupotomy group,electroacupunture group and acupotomy-electroacupunture group(each n=6).The model of the third lumbar transverse process syndrome was established by embedding gelatin sponge into the posterior of the left third lumbar transverse process in all groups except the normal group.After 15 days the acupotomy group and electroacupunture group were given the intervention in bilateral Weizhong(BL40) by acupotomy or electroacupunture respectively,and the acupotomy-electroacupunture group was given the intervention of acupotomy and electroacupunture simultaneously.On the 28th day after modeling,the blood samples were collected from the abdominal aorta of the rabbits for detecting the changes of serum SOD,MDA and T-AOC.The pathological changes of local tissue were observed after HE staining.Results In the model group the activities of serum SOD and T-AOC decreased significantly and MDA level increased significantly with a statistical difference(P0.01,P0.05).The levels of SOD,T0AOC and MDA were not different in the acupotomy group,electroacupunture group and acupotomy-electroacupunture group compared with those in the normal group(P0.05).In the acupotomy group the activities of SOD and T-AOC increased significantly and MDA level decreased significantly compared with those in the model group(P0.05).The activity of SOD increased significantly in the electroacupunture group and acupotomy-electroacupunture group compared with that in the model group(P0.05,P0.01).The level of MDA decreased significantly in the acupotomy-electroacupunture group compared with that in the model group(P0.05).The HE staining showed that there were obvious inflammatory reaction in local tissue in the model group and that were relieved in the acupotomy group,electroacupunture group and acupotomy-electroacupunture group.Conclusion The intervention of acupotomy therapy or(and) electroacupunture can clear off excessive oxygen free radical in the body,prevent the tissue from further injury and improve the recovery of damaged tissue through relieving local inflammatory reactions,increasing the activities of serum SOD and T-AOC,and decreasing serum MDA level.The effect of acupotomy therapy is better than that of electroacupunture.
