HPLC法测定银翘解毒颗粒中连翘苷和牛蒡子苷的含量

Simultaneous determination of Arctiin and Forsythin in Yingqiao Jiedu Granules by HPLC

目的:建立以RP-HPLC法测定银翘解毒颗粒中连翘苷和牛蒡子苷含量的方法。方法:采用Agilent C18(4.6 mm×250 mm,5μm)色谱柱,流动相:乙腈-水(27∶73),检测波长:280 nm,柱温:30℃,流速:1.0 ml/min。结果:连翘苷在0.021 42～0.107 10μg范围内进样量与峰面积呈良好线性关系,平均加样回收率为99.93%,RSD为0.71%;牛蒡子苷在0.836 8～6.694 0μg范围内进样量与峰面积呈良好线性关系,平均加样回收率为99.75%,RSD为0.44%。3批样品中连翘苷和牛蒡子苷的含量分别为0.043 18、1.073 00 mg/g;0.043 06、1.070 00 mg/g;0.043 10、1.071 00 mg/g。结论:本法灵敏、准确,专属性强,重现性好,可作为银翘解毒颗粒中连翘苷和牛蒡子苷的含量测定方法。

Objective： To establish a method for determination of arctiin and forsythin in Yingqiao Jiedu Granules by HPLC. Methods： The analytical column Cj8 （4.6 mm×250mm,5μm） was used. The mobile phase was composed of acetonitrile and water （27：73） at a flow rate of 1.0 ml/min. The wavelength of UV detector was 280 nm and column temperature was 30℃. Results： The linear ranges of arctiin was 0.021 42-0.107 10 μg. The average recovery was 99.93% with a RSD of 0.71%. The linear ranges of forsythin was 0.836 8-6.694 0 μg. The average recovery was 99.75% with a RSD of 0.44%. The content of arctiin and forsythin in three batches of samples were 0.043 18, 1.073 00 mg/g; 0.043 06, 1.070 00 mg/g; 0.043 10, 1.071 00 mg/g. Conclusion： This method is sensitive, accerate, exclusive reproducible. It can be applied to determinate arctiin and forsythin in Yingqiao Jiedu Granules.
