金荞麦对克雷伯杆菌肺炎大鼠心肌损伤中炎症细胞因子表达的影响 被引量:11

摘要 目的研究金荞麦对克雷伯杆菌肺炎大鼠心肌组织损伤的保护作用机制。方法随机将♂SD大鼠分成正常对照组、模型组、金荞麦(6、3、1.5g生药/kg)3个剂量组、左氧氟沙星(25mg/kg)组。计数外周血中白细胞(WBC)和中性粒细胞(PMN),观察肺和心肌组织的病理改变,免疫组化检测大鼠心肌组织中肿瘤坏死因子-α(TNF-α)及细胞黏附因子-1(ICAM-1)蛋白表达。结果与正常对照组比较,模型组大鼠外周血中的WBC和PMN明显升高,肺脏和心肌组织损伤明显,并且模型组大鼠心肌组织中TNF-α和ICAM-1蛋白表达显著增强(P<0.01)。与模型组比较,金荞麦组和左氧氟沙星组大鼠外周血中的WBC和PMN明显降低,心肌组织损伤明显改善,大鼠心肌组织中TNF-α和ICAM-1蛋白表达明显减弱(P<0.05或P<0.01)。结论 TNF-α和ICAM-1蛋白表达上调参与了肺炎大鼠心肌组织的损伤,金荞麦可通过下调其表达来保护肺炎大鼠心肌组织的损伤作用。 Objective To study the mechanism of protective effect of Jinqiaomai(JQM) on heart injury induced by Klebsiella pneumonia in rats. Methods The male SD rats were randomly divided into control group, model group, Jinqiaomai (6,3,1.5 g/kg) three groups , levofloxacin ( 25 mg/kg) group. The white blood cell(WBC) and polymorphonuclear leucocyte ( PMN ) count in peripheral blood were counted . The pathological change of lung and heart were observed . TNF-α and ICAM-1 protein expressions were measured by immunohistochemistry in the cardiac muscle. Results Compared with the control group, the counts of WBC and PMN in peripheral blood were markedly increase in model group and had obvious lung and heart injury, but those of Jinqiaomai and levofloxacin group had less injury. The expressions of TNF-α and ICAM-1 in model group were significantly higher than those in the control group(P 〈0.01),while those of Jinqiaomai and levofloxacin groups were significantly lower than those of model group(P 〈0. 05 or P 〈0. 01 ). Conclusion That heart injury induced by Klebsiella pneumonia is related to TNF-α and ICAM-1. To decrease the excessive expression of TNF-α and ICAM-1 may be the main mechanism of protective effect of Jinqiaomai on heart injury.
