牛血浆FN对人牙髓成纤维细胞骨形成蛋白和碱性磷酸酶分泌和表达的影响 被引量:2

摘要 目的:观察纤维粘连蛋白(FN)与牙髓细胞分化的关系.方法:将传代的人牙髓细胞接种于96孔板,分别用不同浓度牛血浆FN(10,20,40,80μg/ml)作用3天,进行骨形成蛋白(BMP)和碱性磷酸酶(ALP)的免疫组化染色,图象分析仪半定量测定,并测定培养上清液中ALP活性.结果表明:四种浓度的FN组的牙髓细胞BMP和ALP表达无明显差异,FN(20,40μg/ml)组培养上清液中ALP活性较强.结论:FN在牙髓细胞分化过程中的作用是有限的. Aim: To observe the rclationship between fibronectin(FN)and the differentiation of the dental pulp cells.Methods:The human pulp cells at passages 5 were used for the experiments,the cells at the proliferating stage were seeded on the plastic 96-well plates.Four concentrations of FN(10,20,40,80g/ml)were added to the fresh DMEM supplemented with 1%FCS,then the cultivation were continued for 3d,the cells were fixed and then ABC-peroxidase labeling were used to study the intracellular production of alkaline phosphatase(ALP)and bone morphogenetic protein(BMP),ALP activity of culture supernatnant were also measured.Result:There were no signifcant difference on expression of BMP,ALP in FN groups,but FN(20,40g/ml)stimulated ALP activity in supernatant.Conclusion:The role of FN involved in differentiation of dental pulp cells may be limited.
