减肥胶囊对肥胖大鼠脂肪组织瘦素受体后JAK-STAT信号转录系统的影响 被引量:4

摘要 目的:研究减肥胶囊对瘦素受体后信号转录系统的影响,从分子水平探讨减肥胶囊的减肥机制。方法:采用77只普通级SD雄性仔鼠,随机分为空白组10只和高脂饲料组67只,两组体重比较无显著性差异。空白组给予普通饲料,高脂饲料组给予高脂饲料,喂养8周后,选取高脂饲料组大鼠体重高于空白组大鼠平均体重的20%作为肥胖大鼠模型。将复制成功肥胖大鼠随机分为模型对照组、减肥胶囊高剂量组、低剂量组3组。各组均喂普通饲料,高剂量组和低剂量组分别按13.7,6.85 g·kg-1 ig减肥胶囊水煎液(10 mL·kg-1·d-1),空白组和模型对照组ig等体积蒸馏水,共ig 4周。4周后,称重,麻醉大鼠,腹主动脉穿刺取血后,迅速摘取下丘脑和生殖器周围脂肪,采用组织原位杂交法检测下丘脑和脂肪组织酪氨酸蛋白激酶(JAK1),信号转导及转录活化因子(STAT3),信号转导及转录活化因子6(STAT6)基因表达水平。结果:不同浓度的减肥胶囊水煎液能够提升肥胖大鼠下丘脑和脂肪组织中JAK1,STAT3,STAT6基因表达水平,但同时发现,模型对照组下丘脑和脂肪组织中JAK1,STAT3,STAT6基因也处于高水平表达;高剂量组和低剂量组之间存在着一定的不统一性,存在着一定的量效关系。结论:减肥胶囊的作用机制可能是通过对下丘脑和脂肪组织中瘦素受体后信号转录系统的影响而发挥作用,但其机制还需要作进一步的探讨。 Objective: To explore the influence of Jianfei capsule to leptin receptor (LR) behind signal transcription system and the mechanism at Molecular level. Method : The Seventy-seven male conventional Spraguedawleg(SD) rats were divided into control diet group (n = ten) and high fat group (n = sixty-seven) randomly, there is no significant difference on the side of weight(P 〈 0.05). And control diet group were fed with normal diet and high fat diet respectively. After eight weeks, thirty-five rats from high fat diet group were successfully made into obese models . Their body weight were all higher 20% than the average weight in control diet group. They were randomly divided into three groups. Obese model group, high dosage group and low dosage group. All of the groups were replaced by normal diet. Respectively by 13:7,6.85 g· kg^-1, high and low dosage group were given Jianfei capsule decoction (10 mL· kg^-1· d^-1) simultaneously, control group and obese model group were given equivoluminal distilled water. After oral administration for four weeks, all of the rats were anaesthetized, weighted. After adopt blood from aorta abdominalisthe, Hypothalamic and genital fat,liver and heart were collected quickly. Gene expression of JAK1, STAT3, STAT6 of Hypothalamic and adipose tissue were detected by non-radioactive in situ-hybridization. Result:There are the high gene expression of JAK1, STAT3, STAT6 not only in Jianfei capsule but also in obese model group; there is some difference and dose-effect relationship between high dosage group with low dosage group. Third, different concentration Jianfei capsule can improve gene expression of JAKI, STAT3, STAT6 of hypothalamus. Conclusion:The mechanism that Jianfei capsule take effect through its influence to LR behind signal transcription system is still needed to be investigated further.
