荷叶黄酮对油酸孵育的HepG2细胞甘油三酯代谢的影响 被引量:8

摘要 目的研究荷叶黄酮对油酸孵育的HepG2细胞甘油三酯含量的影响以及甘油三酯代谢酶CPT1A活性、蛋白表达的变化。方法以油酸孵育HepG2细胞24h建立肝细胞脂肪变性模型,分组:对照组,模型组,阳性对照组(普伐他汀100mg·mL-1),荷叶黄酮低、中、高剂量治疗组(剂量分别是50、100、200mg·mL-1)。药物孵育24h、48h后测定细胞内甘油三酯含量。测定荷叶黄酮孵育24h后CPT1A酶活性,Westernblot检测CPT1A蛋白表达。结果荷叶黄酮呈时间和剂量依赖性降低模型细胞甘油三酯含量(P<0.05),荷叶黄酮大剂量组治疗24h可以使CPT1A酶活性提高36.9%,荷叶黄酮呈剂量依赖性增加CPT1A蛋白表达(P<0.05)。结论荷叶黄酮可以减轻脂肪变性肝细胞脂滴沉积,降低甘油三酯含量,机制与增强甘油三酯代谢酶CPT1A活性及蛋白表达有关。 Aim To investigate the influence of flavonoids derived from lotus on the content of triglyceride and the regulation of Carnitine palmitoyltransferase 1A(CPT1A)in HepG2 cells.Methods The steatosis liver models were established with oleic acid in vitro.Cells were randomly divided into six groups:normal groups,model groups,pravastatin groups(100 mg·L^-1),flavonoids in lotus groups(50,100,200 mg·L^-1).The level of triglyceride in model cells were measured after 24 h and 48 h treated with flavonoids in lotus.The enzyme activity of CPT1A was measured after 24 h treated with flavonoids in lotus.Western blot was used to measure the protein expression of CPT1A.Results Flavonoids in lotus could effectively reducethe levels of triglyceride in HepG2 cells in time-dependent and dose-dependent manner(P〈0.05).After treatment with 200 mg·L-1 flavonoids in lotus,the activity of CPT1A enzyme increased by 36.9%,and expression of CPT1A was increased dose-dependently.Conclusions Flavonoids in lotus reduce the levels of triglyceride in steatosis liver models in vitro.The changes of CPT1A expression and activity when treated with flavonoids in lotus may play an important role in the regulation of triglyceride in HepG2 cells.
