摘要
为了筛选对木质素有降解能力的菌株,本试验从菜地土壤、动物粪便、青贮饲料中分离筛选出79株产芽孢细菌,初筛获得具有木质素降解能力的菌株20株,经过复筛,筛选出一株具有较高木质素降解能力的菌株T-8,其木质素过氧化物酶酶活为626.67 U/L。对T-8进行形态鉴定、生理生化特征鉴定和16S rDNA序列分析,鉴定该菌株为解淀粉芽孢杆菌(Bacillus amyloliquefaciens),其与B. amyloliquefaciens的相似性高达99.91%。
In order to obtain the strain with lignin-degrading capability,79 spore-forming bacterial strains were isolated and screened from specimen collected from the vegetable field,animal manure and silage feed.Total 20 strains with lignin-degrading capability were obtained through the primary screening.Among them,a strain named T-8 was found with higher activity of degrading lignin than other strains via the secondary screening,and the activity of lignin peroxidase was 626.67 U/L.The morphology characteristics,physiological and biochemical characteristics and 16S rDNA sequence of strain T-8 were further studied.It was finally identified as a Bacillus amyloliquefaciens.The similarity of the 16S rDNA sequences between strain T-8 and the other strains of B.amyloliquefaciens was up to 99.91%.
