丙泊酚预处理对新生猪缺氧\窒息再灌注氧化损伤的影响

摘要 目的:研究丙泊酚对新生猪脑组织缺氧\窒息再灌注氧化损伤的影响。方法:将24只新生猪分为空白对照组(Sham)、缺血缺氧组(HI)和丙泊酚预处理组(Prop-HI)。Prop-HI组(3mg/kg.h)静脉泵注丙泊酚(Sham组和HI组为生理盐水)。2h后HI组和Prop-HI组按文献复制HIE模型。ROSC后第1、2、3和4d行神经行为学变化评分(NBS),观察第5d脑组织病理改变和存活神经元数量,评价丙泊酚能否诱导新生猪产生缺血耐受。检测脑组织ROS含量和血清SOD活性,探讨具体机制。结果:恢复期NBS评分Sham组<Prop-HI组<HI组;壳状核和尾状核存活神经元密度测定HI组<Prop-HI组<Sham组。脑组织ROS含量Sham组<Prop-HI组<HI组,药物预处理后2hSOD活性为Sham组<HI组<Prop-HI组。结论:丙泊酚可诱导新生猪产生缺血耐受,该机制可能与激活过氧化物清除系统有关。
