摘要
以掌叶木[Handeliodendron bodinieri (Lévl.) Rehd.]的叶片、茎段为外植体,在含有不同植物激素的培养基上诱导愈伤组织。结果表明,最佳诱导培养基为MS+2,4-D1.0mg/L+NAA0.5mg/L+6-BA0.5mg/L,最佳增殖培养基为3/4B5+NAA0.5mg/L+6-BA1.0mg/L+VC0.1%。2,4-D与NAA联合使用利于诱导率的提高;适量的6-BA、KT利于愈伤组织质量的改善;6-BA加KT的双因子组合没有单因子的诱导效果显著;采用B5培养基代替MS培养基,且将大量元素降为3/4,外加VC0.1%,并进行暗培养,能有效抑制愈伤组织的褐化。
The leaves and stems of Handeliodendron bodinieri(Lévl.) Rehd were used as explants to induce callus on medium containing different hormones. The results indicated that the best medium for callus induction was MS+2,4-D 1.0 mg/L+NAA 0.5mg/L +6-BA 0.5 mg/L. The optimim medium for enrichment callus was 3/4B5+NAA 0.5 mg/L+6-BA 1.0 mg/L+ VC 0.1%. 2,4-D combined with NAA could increase inductivity. 6-BA or KT in proper quantity could improve the quality of callus; but 6-BA combined with KT showed less effectiveness than a single factor of the two. Changing MS media into B5 media, decreasing major element to 3/4, adding 0.1% antiascorbic acid in the media and darkness culture could inhibit callus browning efficiently.
