组织工程角膜上皮治疗兔角膜缘干细胞缺损模型的实验研究

摘要 采用全板层切除术制作兔眼角膜缘干细胞完全缺损模型,取对侧眼角膜缘组织进行干细胞培养,并以纤维蛋白为基质网架构建组织工程角膜上皮.将实验动物随机分为实验组和对照组,对照组移植不含干细胞的凝胶膜,术后观察1个月.以角膜上皮染色、角膜混浊及新生血管3项指标进行效果评价.结果显示,实验组角膜上皮是一个逐渐修复的过程,角膜水肿、炎性反应逐渐减轻,新生血管明显减少.由此得出,以纤维凝胶膜为一种新型的组织工程材料制备的组织工程角膜上皮经移植术后证实可修复角膜上皮的损伤. An ocular surface defect was created in the right eyes of the experiment rabbits by a lamellar keratectomy extending 1 mm outside the limbus. The stem cells were isolated from a limbal biopsy specimen from the left eyes and cultured. The cells were combined with fibrin gel membrane to construct bioengineered corneal epithelium. Rabbits were divided into 2 groups at random after cytologic verification of limbal stem cell deficiency. Culture skin epidermal stem cells and keratoeytes were in good growth conditions. One month after transplantation, the cornea wounds were healed and the rabbit cornea became transparent. Tissue bioengineered cornea epitheliums have no immunological rejection, while they have good histoeompatibility, which will be useful transplant material for treating limbal stem cell deficiency.
