摘要 目的:探讨可能影响皖南地区五步蛇毒蛋白C激活物(PCA)酰胺酶活性的实验因素。方法:分别采用发色底物法和APTT法检测皖南地区五步蛇毒PCA在不同实验条件下的酰胺酶活性。结果:当实验温度为37℃时该PCA活性最稳定,而温度升高或降低均会抑制其酰胺酶活性(P<0.05);该PCA在pH值为7.0时活性最稳定,偏酸或偏碱则均会明显抑制其活性(P<0.05);但一定浓度范围内的Na+、K+、枸橼酸钠等对其活性无明显影响(P>0.05)。结论:五步蛇毒PCA是一种效价较高、影响因素较少的蛋白C活化试剂。 Objective:To investigate the potential influential factors in certain experimental conditions on amidase activities of protein C activator(PCA)isolated from venom of Agkistrodon acutus(AAV)from southern Anhui province.Methods:Amidase activities of PCA from AAV were measured in diverse experimental conditions by respective method of chromogenic substrate and active partial thromboplastin time(APTT).Results:The amidase activities of PCA remained most stable at 37 ℃,and yet,the activities were found inhibited with increase or decrease of the temperature(P0.05).When the pH value of PCA kept at 7.0,the activities were checked significantly under either acidic or alkali conditions(P0.05),whereas the activities were free from effect with specific concentration regarding variation of the substrate Na+,K+ and sodium citrate(P0.05).Conclusion:PCA from AAV is a favorable protein C-activating reagent that has high potency and produces less influence on the experiment.
