摘要 目的研究微囊化人胎肾上腺皮质细胞的制备及其功能,并进行形态学观察。方法将人胎肾上腺皮质制成单细胞,再用具有良好生物相容性的海藻酸钠聚赖氨酸海藻酸钠(APA)微囊包裹,制成微囊化肾上腺皮质细胞。将分离的肾上腺皮质细胞和微囊化肾上腺皮质细胞分组进行培养,测定培养22、46、70小时的培养液中的皮质醇含量,在取培养液的同时加入含促肾上腺皮质激素(125ng/L)的培养液刺激培养2小时,即培养中的24、48、72小时再取培养液测定皮质醇含量。结果形态学观察发现机械法分离的肾上腺皮质细胞存活率为92%,制备成微囊后的肾上腺皮质细胞存活率为83%;微囊化肾上腺皮质细胞形态完整,肾上腺皮质细胞均匀地分布在APA微囊内,微囊表面光滑完整;两个培养组的皮质醇含量变化的差异无显著性(P>0.05),微囊对肾上腺皮质细胞的活性无显著影响。 Objective To study the preparation of microencapsulated human embryo adrenal cortical cells and their morphology and functions. Methods Single cell from human embryo adrenal cortex was made, enwrapped with better biological consistent alginate polyLlysine alginate (APA) and microencapsulated adrenal cortical cells were developed. The isolated adrenal cortical cells (group A) and microencapsulated adrenal cortical cells (group B) were cultured separately. The cortisol levels in the culture fluid at 22nd, 46th, 70th h were determined in the two groups, respectively. After ACTH (125ng/L) addition into the culture fluid for stimulating culture for 2h, the cortisol levels in the culture fluid at 24th, 48th and 72nd h were measured again. Results Morphologically, the total cell survival rate of the embryonic adrenal cells was 92% by using mechanical methods, while the survival of microencapsulated cells was 83%. Microencapsulated adrenal cells' shape was intact, evenly distributing in APA. The surface of microencaps was smooth. The cortisol levels in two groups had no significant difference. Microencapsulation had no obvious influence on adrenal cell activity. Conclusion The microencapsulation had no harmful to the survival and secretion function of adrenal cells.
