聚合酶链反应在器官移植受者人类疱疹病毒6型感染检测中的应用

摘要 目的探讨器官移植受者人类疱疹病毒6型(HHV6)感染的诊断方法。方法应用聚合酶链反应(PCR)技术,检测31例器官移植受者术前1周、术后16天及10例移植术后发热时的外周血单个核细胞(PBMCs)中的HHV6DNA。结果术前7例和术后19例的PBMCs中检测出HHV6DNA,10例发热患者中8例HHV6DNA阳性。对10例发热患者用阿昔洛韦治疗,8例症状得到控制,2例死亡。结论应用PCR技术检测HHV6感染具有简便、快速、准确、特异性高等优点。 Objective To explore the diagnosis of human herpesvirus 6 (HHV6) infection in organ transplant recipients. Methods HHV6 DNA in peripheral blood mononuclear cells (PBMCs) in 31 organ transplant recipients was detected 1 week before transplantation and 16 days after transplantation by using polymerase chain reaction (PCR). Results HHV6 DNA in PBMCs was detectable in 19 cases after operation and 7 before operation among the 31 cases respectively, and 8 out of 10 recipients with fever after operation were positive for HHV6 DNA. The 10 recipients with fever were treated by antivirus chemotherapy (acyclovir), 8 out of 10 were effective. Conclusion PCR amplification is a suitable method to diagnose and monitor HHV6 infection. The method is simple, rapid, sensitive (sensitivity 97%), and specific (specificity 98%).
