亮叶杨桐叶总类黄酮的提取工艺优化及对肺腺癌细胞A549生长的影响 被引量:5

Optimization of Total Flavonoids Extraction from the Leaves of Adinandra nitida and Their Inhibitory Effect on Lung Cancer A549 Cells

摘要 采用响应面法对亮叶杨桐叶中类黄酮物质的提取工艺条件进行优化。结果表明,提取时间10min、乙醇体积分数63%、提取温度70℃、比料液1:20时,亮叶杨桐叶中总类黄酮提取率为(95.58±0.49)%。采用MTT法对亮叶杨桐叶类黄酮抗肺腺癌细胞A549生长的活性进行研究,结果显示亮叶杨桐叶类黄酮对肺腺癌细胞A549具有良好的抑制作用,并呈现显著的时间效应和剂量效应关系。 Adinandra nitida is a traditional Chinese herb for health drink, of which leaves are rich in flavonoids. The optimization of total flavonoids extraction from the leaves ofAdinandra nitida with ethanol as the extraction solvent was conducted in this work, which was also aimed at examining if the total flavonoids from the leaves of Adinandra nitida have the ability to inhibit the proliferation of lung cancer A549 cells. The optimal extraction conditions of total flavonoids from Adinandra nitida leaves were determined by response surface methodology to be: extraction time, 10 min; ethanol concentration, 63%, extraction temperature, 70 ℃; and material/liquid ratio, 1:20. The extraction rate of total flavonoids was (95.58 ± 0.49) % under the optimal extraction conditions. The inhibitory effect of the total flavonoids from the leaves ofAdinandra nitida on the proliferation of lung cancer A549 cells was evaluated by MTT assay. The total flavonoids could significantly inhibit the proliferation of lung cancer A549 cells in significant time-dependent and dose-dependent manners.
