摘要
用原核表达的汉坦病毒76118株核蛋白为夹心抗原,组合抗人μ链多克隆抗体和酶标抗汉坦病毒单克隆抗体,建立了IgM捕捉法ELISA,用以检测610份疑似患者血清,并与免疫荧光法(IFA)相比较。结果本方法与IFA的阳性符合率9410%、阴性符合率9283%、总符合率9361%,检出率在发病5日内明显高于IFA,6至30日无区别,30天后开始下降,检出病例的月分布与发病规律相符。表明重组核蛋白抗原的IgM捕捉ELISA能够准确早期诊断HFRS。
For studying the value of IgM capturing ELISA with recombinant antigen to the serodiagnosis of HFRS,The ELISA was established with antigen expressed from 76118 strain S genome segment in E.coli,combining with polyclonal Ab against human μ chain and HRP labeled monoclonal Ab against Hantavirus.The method was used to detect 610 sera of suspected HFRS pstients,and the results were compared to that of Indirect Fluorescent Assyay.The coincidences of ELISA with IFA were 94 10% for positive sera,92 83% for negative,and 93 61% for general;the sensitivities of the ELISA were apparently higher than that of the IFA for the patients that within 5 days ill length ,equal for that with 6 to 30 days ill length,and lower for that with 1month or longer ill length;the month distribution of porsitive found was accod with the infection rule.
