摘要
目的研究环孢素A(cyclosporin A,CsA)对脂多糖(lipopolysaccharide,LPS)诱导的小鼠早期炎症反应的影响。方法小鼠腹腔内注射LPS诱导炎症反应,静脉注射不同浓度的CsA。6h后取血清,使用ELISA方法观察炎性因子IFN-γ和IL-6的含量;取脾细胞,使用流式细胞仪观察CD4+和CD8+T细胞表面的活化分子CD69表达;并使用细胞内细胞因子染色的方法,观察CD4+和CD8+T细胞IFN-γ和IL-4的产生水平。结果 CsA可以明显降低LPS诱导的小鼠血清中IFN-γ的水平,抑制CD4+和CD8+T细胞表面CD69分子的表达,抑制T细胞IFN-γ和IL-4的产生,均表现为剂量依赖性抑制关系。结论 CsA剂量依赖性抑制LPS诱导的小鼠早期炎症反应。
We aimed to evaluate the effects of cyclosporin A(CsA) on early inflammatory reaction induced by LPS.Early inflammatory reaction was induced in mice with intraperitoneal injection of LPS to mice to induce,and then different doses of CsA were intravenous injected to these mice.Six hours later,serum were obtained,and the levels of IL-6 and IFN-γ were detected by ELISA;splenocytes were prepared,and the expression of CD69 on the surface of CD4+ and CD8+ T cells were assayed by Flow Cytometric analysis.The frequency of IFN-γ and IL-4 producing cells in CD4+ and CD8+ T cells were examined by the means of intracellular cytokines staining.We found that CsA decreased INF-γ levels in LPS induced mice,apparently.In the same time,CsA inhibited the expression of CD69 on the surface of CD4+ and CD8+ T cells,as well as inhibited the production of IFN-γ and IL-4.All the inhibiting effects were in a dose dependent manner.As the conclusion,CsA could inhibit LPS induced early inflammatory reaction in mice in a dose dependent manner.
