骨形态发生蛋白9对人乳腺癌MDA-MB-231细胞增殖与凋亡的作用研究 被引量:4

摘要 目的:研究骨形态发生蛋白9(Bone morphogenetic protein 9,BMP9)对乳腺癌细胞MDA-MB-231增殖、周期及凋亡的影响。方法:半定量RT-PCR法检测高转移性乳腺癌细胞MDA-MB-231和低转移性乳腺癌细胞MCF-7中BMP9的表达,用人BMP9重组腺病毒(AdBMP9)感染MDA-MB-231细胞作为实验组,含GFP的空载腺病毒(AdGFP)感染MDA-MB-231细胞作为对照组,MTT法观察细胞增殖的变化,流式细胞仪分析周期及凋亡的变化。结果:半定量RT-PCR显示,乳腺癌细胞MDA-MB-231中未检测到BMP9 mRNA的表达;MTT结果显示,AdBMP9处理可引起MDA-MB-231细胞增殖抑制,第5 d MDA-MB-231/BMP9组的细胞增殖率(0.8860±0.0532)显著低于MDA-MB-231/GFP组(1.2240±0.1031)(P<0.05);流式细胞仪分析结果显示,腺病毒感染后第2 d和第3 d,MDA-MB-231/BMP9组细胞周期各相发生变化,G2/M期细胞明显增加,分别为MDA-MB-231/GFP组的3.2倍和2.4倍(P<0.05);同时BMP9可诱发细胞凋亡,腺病毒感染后第3 d MDA-MB-231/BMP9组细胞凋亡百分率(31.55%±8.26%)明显高于MDA-MB-231/GFP组(3.80%±0.46%)(P<0.05)。结论:提示BMP9可通过阻滞细胞周期进程和诱导细胞凋亡来抑制乳腺癌细胞MDA-MB-231的增殖。 Objective:To investigate the effects of bone morphogenetic protein 9 on the proliferation, cell cycle and apoptosis of breast cancer cell line MDA-MB-231.Methods:The expression of BMP9 in high-metastatic cell line MDA-MB-231 and low-metastatic cell line MCF-7 were detected by Semi-quantitative PCR respectively,MDA-MB-231 were transfected with AdBMP9(as experimental groups) and AdGFP(as control groups) respectively,Then the ability of cell proliferation was detected by MTT,cell cycle and apoptosis were detected by flow cytometry.Results:The BMP9 mRNA was not detected in human breast cancer cell line MDA-MB-231.After transfection with AdBMP9,the MDA-MB-231 cells showed an inhibited proliferation,On the fifth day,the growth rate of experimental groups(0.886 0±0.053 2)was significantly lower than that of control groups(1.224 0±0.103 1)(P0.05);The results of flow cytometry showed that,BMP9 could induce the cell cycle arrest in MDA-MB-231,On the second day and third day after transfection,the cell number of G2/M phase increased significantly,which were 3.2 fold and 2.4 fold that of control groups respectively;It's also found that after 72h treatment,the apoptosis rate of experimental groups(31.55%±8.26%) was significantly higher than that of control group(3.80%±0.46%)(P0.05).Conclusion:These data demonstrated that BMP9 could inhibit the proliferation of MDA-MB-231 by blocking cell cycle and inducing apoptosis.
