乌司他丁对A549细胞在缺氧条件下ENaC-α表达影响的研究 被引量:1

摘要 目的:研究乌司他丁对缺氧A549细胞ENaC-α mRNA和蛋白表达水平的影响。方法:将A549细胞分组为缺氧对照组和乌司他丁组,分别在3%O2和0%O2的缺氧条件下,用1 250、2 500、5 000 U/ml浓度乌司他丁作用A549细胞,用RT-PCR半定量ENaC-α mRNA表达量,用Western blot半定量ENaC-α蛋白浓度。结果:在不同缺氧程度下,乌司他丁组的ENaC-α mRNA和蛋白表达均高于对照组,呈浓度依赖方式。结论:乌司他丁能够对抗缺氧对ENaC-α表达的抑制作用,提示乌司他丁在急性肺损伤(Acute lung injury,ALI)的治疗过程中具有缓解肺水肿、减轻肺损伤的作用。 Objective:To study the effect of Ulinastatin on ENaC-α mRNA and protein of human alveolar epithelial A549 cells which are treated with hypoxia.Methods:A549 cells were divided into two groups,hypoxia group and ulinastatin group.A549 cells were treated with Ulinastatin at the doses of 1 250, 2 500 U/ml and 5 000 U/ml for 48 h when cotreated with 3% O2 or 0% O2.The mRNA and protein levels of ENaC-α were determined by semiquantitative RT-PCR and Western blot.Results:Incubation of A549 with 1 250 U/ml,2 500 U/ml and 5 000 U/ml of Ulinastatin for 48 h could significantly upregulate the expression of ENaC-α in a concentration-dependent manner.Conclusion:Ulinastatin could reverse the inhibitory effect of hypoxia on the expression of ENaC-α,which suggests that ulinastatin can promote lung fluid clearance and pulmonary edema reduction.
