摘要
目的:探讨建立C57BL/6j小鼠人神经母细胞瘤移植瘤模型可行性。方法:取含1×107个对数生长期SK-N-SH人神经母细胞瘤细胞悬液0.2 ml,接种于C57BL/6j系小鼠右前肢腋窝皮下,观察小鼠成瘤率、相对肿瘤体积(Relative tumor volume,RTV)、相对肿瘤增长速率(K)、相对肿瘤倍增时间(Td)、存活时间及移植瘤病理学情况。结果:C57BL/6j小鼠在3、7 d、3周、4、5、6、7周各年龄段持续成瘤率分别为:100%、100%、90%、70%、60%、0%、0%。成瘤后3~5周内,3周与4周及5周龄接种的肿瘤RTV、K、Td及C57BL/6j存活时间均有差异(P<0.05)。结论:在C57BL/6j小鼠体内成功建立人类异种移植瘤模型。不同年龄段接种的成瘤率有差异,可以根据后期研究目的选择接种建模时间。
Objective:To investigate the feasibility of human neuroblastoma xenograft model in C57BL/6j mice.Methods:SK-N-SH cells were collected and condensed as 1×107/ml during the logarithmic growth phase,0.2 ml injected subcutaneously into C57BL/6j mice.The tumor generation rate,relative tumor volume(RTV),relative growth rate(K),relative doubling time(Td),survival time and pathology were tested.Results:The continuing tumor generation rate of C57BL/6j mice was 100%(10/10) in 3 day,100%(10/10) in 7 day,90%(9/10) in 3 week and 70%(7/10) in 4 week,60%(6/10) in 5 week,0%(0/10) in 6 week and 0%(0/10) in 7 week respectively.There were differences in RTV,K, Td,and survival time(P0.05) when comparing 3 week C57BL/6j mice with 4 week and 5 week ones. Conclusion:Human neuroblastoma xenograft model is successfully established in the immunocompetent C57BL/6j mice.There is significant difference between the inoculated ages in the tumor generation rate,and time-points of model inoculation can be selected according to later research purposes.
