LPS刺激SD大鼠胆管上皮细胞表型转化的动物实验研究 被引量:3

摘要 目的:探讨上皮标志物上皮性钙粘附蛋白(E-cadherhin)和间叶标志物波形蛋白(Vimentin)在脂多糖(Lipopolysacoharides,LPS)诱导的SD大鼠胆管上皮细胞中的表达及其意义。方法:选用SD大鼠作为研究对象,用LPS(2μg/ml)剌激其胆管,用免疫组化方法检测E-cadherin和Vimentin在胆管细胞中的表达情况。结果:SD大鼠胆管上皮细胞经LPS剌激后,E-cadherin蛋白表达逐渐缺失(P<0.05),而Vimentin蛋白出现阳性表达(P<0.05)。结论:LPS剌激可使SD大鼠胆管上皮细胞发生上皮细胞间质转型(Epithelial-mesenchymal transitions,EMT)。 Objective:To explore the expression and importance of epithelial marker E-cadherhin and mesenehymal marker Vimentin in the bile duct epithelial cells induced by LPS.Methods:Epithelia of intrahepatic bile ducts of SD rats were chosen as research subjects,and put into LPS(2μg/ml). Immunohistochemical technique was applied to detect both the markers of epithelial E-cadherin and mesenehymal Vimentin.Results:E-cadherin gradually reduced(P0.05)and Vimentin acquired masculine expression after induction(P0.05).Conclusion:Epithelial-mesenehymal transitions happened in the bile duct epithelial cells inducted by LPS.
