艳山姜果实挥发油对离体家兔胸主动脉条收缩性能的影响 被引量:7

Effects of volatile oils from fructus Alpiniae zerumbet on the contraction of rabbit thoracic aorta in vitro

摘要 目的:研究艳山姜果实挥发油对兔离体胸主动脉血管条收缩性能的影响,为充分利用艳山姜果实资源提供实验依据。方法:采用家兔离体胸主动脉血管条,以不同浓度去甲肾上腺素(NE)、氯化钾为刺激剂,观察艳山姜果实挥发油对其量效曲线的影响;以NE为血管收缩刺激剂,观察艳山姜果实挥发油对内钙释放与外钙内流的影响。结果:艳山姜果实挥发油使NE与氯化钾的量效曲线明显右移,最大反应性降低,对氯化钾量效曲线的抑制作用强于NE;在无钙Kerb′s液中,艳山姜果实挥发油浓度依赖性抑制NE诱导的内钙释放所致的血管收缩反应,对复钙后NE所致的血管持续收缩具有抑制作用的趋势。结论:艳山姜果实挥发油改善兔离体主动脉条的收缩作用与钙离子拮抗有关,且对主动脉依内钙性收缩的敏感性大于依外钙性收缩;对电压依赖钙通道的拮抗作用强于受体操纵的钙通道。 OBJECTIVE To investigate the effects of volatile oils from fructus Alpiniae zerumbet (VOFAZ) on contraction of rabbit thoracic aorta in vitro, to offer the experimental basis for taking full advantage of fructus Alpiniae zerumbet. METH- ODS The effects of VOFAZ on the concentration-response curves of noradrenalin and high potassium were observed on isotonic contractions of The thoracic aorta strips in vitro. The procedure of free-Ca2+Kerb's and Ca2+ addition was designed to indirectly investigate the effects of VOFAZ on intracellular free Ca2 + stimulated by NE. RESULTS VOFAZ shifted the concentra- tion-response curves of NA and KC1, and the maximum responses were decreased. In Ca2+-free medium, VOFAZ concentra- tion-dependemly and significantly inhibited vasoconstriction resulting from Ca2+ release induced by NE and it slightly inhibited the long-lasting contraction induced by addition of Ca2+. CONCLUSION VOFAZ had significant relaxant effect on the contraction of thoracic aorta strips. The effect may be due to the inhibition of the calcium channel of aortic smooth muscle. VOFAZ showed relatively less effect on the extracellular Ca2+ -dependent contraction induced by Ca2+ than that on the intraceilular Ca〉 dependent contraction, and more against effects on potential dependent Ca2+ channel than receptors operated channel.
