正交试验法优选普通针毛蕨地上部位总黄酮提取纯化工艺

摘要 目的:优选普通针毛蕨地上部位总黄酮提取和纯化工艺条件。方法:以总黄酮提取率为考查指标,液料比、提取次数、提取时间、乙醇浓度为考察因素,选用L9(34)表对提取工艺进行正交试验。以总黄酮提取率、总黄酮纯度和主要活性成分原芹菜素(protoapigenone)物质为考查指标,用高效液相色谱法测定原芹菜素含量,以加碱水量,加碱水次数,碱水pH值,加酸后pH为影响因素,选用L9(34)表对纯化工艺进行正交试验。结果:普通针毛蕨总黄酮提取最佳工艺为20倍量75%乙醇提取3次,每次1.5h。纯化条件为药材醇提浸膏加2倍量pH9的氢氧化钠溶液,提取2次,提取液用浓盐酸调节为pH1。结论:优选工艺获得的总黄酮提取率,纯度和原芹菜素含量较高。本方法合理,便于生产。 OBJECTIVE To optimize the conditions of extraction and purification processes for the total flavonoids of the aerial parts of Macrothelypteris torresiana. METHODS Orthogonal test was used for studying the effects of alcohol concentration, volume, extraction time and the number of times on the content of total flavonoids, and the effects of alkali extraction's pH, volume, the number of times and acid precipitation's pH on the content of protoapigenone, purity quotient and extraction rates. RESULTS The optimal extraction process is as follow: 20 times of 75% alcohol, 3 times of extraction and each for 1.5 hours. The optimal purification process is as follow: 20 times of pH 9 alkali extraction, 2 times of extraction and pill acid pre- cipitatiork CONCLUSION The extraction and purification processes are reasonable and fit for industrialism.
