新生猪胰岛细胞微囊体内外分泌功能观察

Study on the functional ability of microencapsulation of neonatal pig islet in vivo and in vitro culture

摘要 目的:比较以聚鸟氨酸、聚赖氨酸为囊材制备的新生猪胰岛细胞微囊的体内、体外分泌功能差异。方法:于高压静电场中用聚电解质络合法分别制备包裹新生猪胰岛细胞的海藻酸钠-聚赖氨酸-海藻酸钠(APA)与海藻酸钠-聚鸟氨酸-海藻酸钠微囊(ALA),行体外培养及糖尿病大鼠腹腔内移植。结果:2组新生猪胰岛细胞微囊的培养液中胰岛素水平均以第3天为最高,组间差异无显著性;胰岛素刺激释放试验组间差异无显著性;体内移植后,ALA组较APA组降血糖作用稍强。结论:APA组、ALA组体外分泌功能相似,体内移植后ALA组降糖作用略强。 OBJECTIVE To observe and compare the functions of two types of microencapsulated neonatal pig pancreatic islets in vivo and in vitro. METHODS The ability of secreting insulin and glucose-stimulated insulin release between the alginicpolyornithine-alginic microencapsuted neonatal pig islet and alginic-polylysine-alginic microencapsuted neonatal pig islet in vivo and in vitro culture were compared. RESULTS The insulin levels in cultures of the two types of microencapsulated neonatal pig pancreatic islets were highest in the 3rd day, but there was no significant difference between that of the two types; there was also no significant difference in glucose-stimulated insulin release between the two types; however , the ability of decrease in plasma glucose level of ALA type was slightly stronger than that of APA type after transplantation. CONCLUSION Both islets had good biological activity in vitro culture, but the alginic polyornithine-alginic microencapsule group had better effects on plasma glucose levels of STUD rats than the Alginic-polylysine-alginic microencapsule's after transplantation. Aiginic polyornithine alginic microencapsule and their making technique have no obvious influence on neither islets cell activity nor their secretion.
