摘要
本研究设计了1对特异性引物,采用PCR-RFLP法对5个家兔群体FGF5基因部分外显子1的序列进行分析,结果显示:FGF5基因外显子1发现了2个等位基因、3种基因型,220-222位点存在TCT缺失。所有的群体均处于哈代-温伯格平衡,且在所选的5个家兔群体的FGF5基因外显子1中,A1等位基因均为优势等位基因,獭兔群体表现为中度多态,肉兔群体表现为低度多态,毛兔未检测到多态。皖系长毛兔与其他家兔群体之间分布差异极显著(P<0.01),九疑山兔与有色獭兔之间分布差异显著(P<0.05),其他群体间不存在分布差异(P>0.05)。
Part of Exon 1 of FGF5 gene in 5 rabbit populations were scanned by PCR-RFLP method with 1 specific primer.The results showed that 3 genotypes and 2 alleles were found at exon 1 of FGF5 loci,there was a TCT Insertion/deletion mutation at 220-222 position.The distribution of these genotypes in all populations was consistent with the Hardy-Weinberg equilibrium,A1 allele was the preponderant allele,Rex rabbit showed moderate polymorphism,meat rabbit showed low polymorphism,Wan line angora rabbit showed no polymorphism.The distribution of these genotypes was very significant(P0.01) between Wan line angora rabbit and all the other rabbit populations,and it was significant(P 0.05) between Jiuyishan rabbit and chinchilla rex rabhit while it showed no significant from one to each other rabbit populations.This study suggested a theoretical reference for the breeding of rabbit wool quality whether FGF5 gene could be a molecular marker or not.
