大蒜素对慢性气道炎症大鼠上皮细胞中NRF2与γ-GCS表达的影响

摘要 目的研究大蒜素对慢性气道炎症大鼠上皮细胞中NRF2与γ-GCS表达的影响。方法将30只SD大鼠随机分为正常对照组、模型组和大蒜素治疗组,每组10只。模型组及大蒜素治疗组用两次气管内滴入脂多糖和每日熏香烟法制成大鼠慢性气道炎症模型,大蒜素治疗组在模型制作成功后腹腔注射大蒜素注射液。通过肺组织HE观察气道炎症改变,支气管肺泡灌洗(BALF)细胞记数并分类,逆转录-聚合酶链反应(RT-PCR)检测NRF2mRNA及γ-GCSmRNA表达,检测肺组织GSH、ROS、T-AOC含量。结果大蒜素治疗组气道炎症浸润较模型组明显减轻,大蒜素治疗组BALF中中性粒细胞数目较模型组降低(P<0.05);模型组的γ-GCSmRNA及NRF2mRNA表达高于大蒜素治疗组(P<0.01);大蒜素治疗组中GSH含量较模型组减少,ROS含量降低,T-AOC上升(P<0.01)。结论大蒜素可抑制被动吸烟大鼠肺部炎症。 Objective To investigate the effect of allicin on nuclear respiratory factor-2(NRF2) and γ-glutamylcysteine synthetase(γ-GCS) on chronic airway inflammation rat. Methods 30 Sprague-Dawley rats were randomized into 3 groups:model group,control group and created by allicin group.Chronic airway inflammation rat model was established by twice intratracheal instillation of lipopolysaccharide(LPS) and exposured to cigarette smoke daily in control group and created by allicin group.Intraperitoneal inject allicin when model be built succed in model group and created by allicin group.Observed the change of airway inflammation by HE in lung tissue.The total cell count and the proportion of inflammatory cells in BALF were measured. The methods of RT-PCR were used for detection of mRNA expressions of NRF2 and γ-GCS.The levels of reduced glutathione(GSH),reactive oxygen species(ROS),total antioxidative capacity(T-AOC) in lung tissue. Results The airway inflammation of created by allicin group relieved.The proportion of neutrophile granulocyte in BALF concentration of the lung tissue in created by allicin group were lower than those in model group(P0.05). The levels of NRF2mRNA and γ-GCSmRNA in model group were higher than created by allicin group.(P0.01).The levels of the GSH and ROS were decreased in created by allicin group.The level of T-AOC was increased in created by allicin group(P0.01). Conclusion Allicin can depress the airway inflammation of rat.
