RP-HPLC法测定新疆不同产地药桑叶中芦丁、异槲皮苷的含量 被引量:6

摘要 目的考察新疆不同产地、不同采摘时间药桑叶中芦丁、异槲皮苷的含量变化。方法采用反相高效液相色谱法(RP-HPLC),以大连依利特Hypersil ODS(150mm×4.6mm,5μm)为色谱柱,进样量20μl,流动相为乙腈(A)-水(B)-0.2%磷酸溶液(pH=4.8)(C),梯度洗脱[0min,A-C(18∶82);4min,A-C(21∶79);10min,A-C(8∶92)],流速1.0ml/min,检测波长为358nm,柱温为25℃。结果不同产地、不同采摘时间的药桑叶中芦丁和异槲皮苷的含量均有一定的差异,喀什地区、5~6月份二者含量较高。结论药桑叶中芦丁和异槲皮苷的含量与产地和采摘季节等有关,尤其以季节的影响大。 Objective To investigate the content difference of rutin and isoquercitrin in drug mulberry leaves from different areas in Xinjiang.Methods Reversed phase high performance liquid chromatography(RP-HPLC)with Yilite Hypersil ODS(150 mm×4.6 mm,5 μm)chromatographic column was used.The sample size was 20 μl,the mobile phase was acetonitrile(A)-water(B)-0.2 % phosphoric acid(C)with gradient elution [0 min,A-C(18∶82);4 min,A-C(21∶79);10 min,A-C(8∶92)].The flow rate was 1.0 ml/min,the detection wavelength was at 358 nm,and the column temperature was 25℃.Results The contents of two components changed at some degree in drug mulberry leaves from different plant sites,while the contents varied obviously among drug mulberry leaves from different collection time and different varieties.Conclusion The contents of the two components are influenced by collected time and plant site,espacially by collected time.Both rutin content and isoguercitrin content are higher in the mulberry leaves picked in kashi,in June and July.
