新疆石榴皮乙酸乙酯提取部位的制备和初步鉴定

Preparation and initial identification of acetic acid extract of Xinjiang pomegranate peel

目的对新疆石榴皮的乙酸乙酯提取部位进行制备和鉴定。方法用乙酸乙酯回流提取制备浸膏,用化学显色法和薄层色谱法对提取物进行鉴定。结果最佳提取条件:固液比1∶20,提取温度80℃,提取次数3次,提取时间分别为3、2、1.5h,出膏率4.3%;系统预试验结果显示,提取物可能含有黄酮类、生物碱、有机酸、酚类和挥发油等成分;以展开剂为正丁醇-醋酸-水(4∶1∶8)的硅胶薄层色谱板上显示4个鲜明的薄层斑点(Rf=0.3～0.5)。结论为新疆石榴皮乙酸乙酯提取部位的制备、化学成分分析、质量标准研究及抗糖尿病产品开发奠定重要基础。

Objective To prepare and identify initially the acetic acid extract of Xinjiang pomegranate peel.Methods Acetic acid extract was prepared from pomegranate peel by reflux-heating method;chemical and chromatographic（TLC）identification were performed.Results The optimal extraction condition was：solid/liquid ratio 1∶20,80℃/3 h,extraction yield 4.3%;the chemical identification showed that the extract possible contained flavonoid,alkaloids,phenolic compounds,organic acid and volatil oil;the result of TLC,n-butyl alcohol-acetic acid-water（4∶1∶8）as the developing solvent,shows 4 clear TLC spots（Rf=0.3~0.5）.Conclusion This study could provide a fundamental basis for preparation,structure analysis,quality control and pharmacological study of acetic acid extract of pomegranate peel,for further development of anti-diabetic new agent.
