耐力运动对ApoE基因缺陷小鼠肝X受体a和三磷酸腺苷结合盒转运体A1基因表达的影响 被引量:1

Effect of Endurance Exercise on LXRa andABCA1 gene expression in Apoe-Deficient Mice and Its Intervention from Atherosclerotisis

摘要 目的:观察耐力运动对ApoE基因敲除(ApoE-/-)小鼠肝脏肝X受体a(LXR-a)和三磷酸腺苷结合盒转运体A1(ABCA1)基因表达的影响及其在抗动脉粥样硬化中的作用。方法:将24只8周龄ApoE基因敲除小鼠随机分为AS模型组和运动干预组,每组12只,运动组进行14周跑台训练,实验持续14周后,采用普通PCR法测定肝脏LXR-a及ABCA1mRNA表达;采用HE染色法观察主动脉根部的病理变化。结果:运动组与安静组相比,LXR-a和ABCA1mRNA表达上调(P<0.05),主动脉斑块及内膜损伤程度较对照组减轻。结论:耐力运动通过增加LXR-a和ABCA1mRNA的表达,而发挥抗动脉粥样硬化的作用。 Objective:To investigate the effects of endurance training on expressions of LXRα and ABCA1in in ApoE-deficient mice and to explore the relation of LXRα with Atherosclerosis.Eighteen 8-week old ApoE-deficient mice were selected and divided into atherosclerosis model group and exercise group randomly.There were twelve mice in each group.Mice of exercise group were trained on treadmill for fourteen weeks.The LXRα and ABCA1mRNA of liver were determined by reverse transcriptase polymerase chain reaction(PCR).The pathological change were determined by HE staining.Results:The expression of LXRα and ABCA1mRNA were significantly increased(P0.05)in ApoE-deficient mice with endurance exercise.The degree of lesion in aorta wall and tunica intima was lighter in ApoE-deficient mice with endurance exercise.Conclusions:The present study demonstrated that endurance exercise may contribute to its anti-atherosclerosis function by significantly increasing LXRα and ABCA1 mRNA.
