P38信号通路在Aβ介导的PC12细胞损伤中的作用

P38 signaling pathway participates in PC12 cell injury induced by Aβ

摘要 目的探讨P38信号通路在Aβ介导的PC12细胞损伤中的作用。方法体外培养PC12细胞,不同浓度的Aβ处理PC12细胞。用MTT法检测Aβ对PC12细胞活力的影响,免疫印迹法检测P38通路活化水平,并观察P38通路抑制剂SB203580对细胞活力的影响。结果 Aβ处理后,PC12细胞活力随Aβ浓度的增高而逐渐下降(P<0.05);Aβ处理后P38表达水平从4h开始升高,12h达到高峰,并一直持续到24h;SB203580预处理能够明显抑制P38信号通路活化,并对PC12细胞起到保护作用。结论 P38信号通路活化参与Aβ介导的PC12细胞损伤。 Objective To investigate the role of P38 signaling pathway in PC12 cell injury induced by Aβ.Methods PC12 cells were treated with Aβ.Cell viability of PC12 cells after exposure to Aβ was detected by MTT method.Western blotting was used to verify the activation of Aβ.We also investigated the role of SB203580,an specific inhibitor of P38 on Aβ-induced P38 signaling pathway activation and cell viability.Results After Aβ treatment,the viability of PC12 cells declined as the concentration of Aβ increase.After exposure to Aβ,the activation of P38 was observed 4 h after Aβ exposure,reached the peak at 12 h,and lasted at lease 24 h.SB203580 inhibited P38 activation induced by Aβ,and attenuated cell injury.Conclusions P38 signaling pathway participates in Aβ-induced cell cycle injury.
