摘要 目的:研究电针足三里、曲池对脑缺血再灌注损伤大鼠下丘脑β-内啡肽(β-EP)、谷氨酸(Glu)蛋白表达的影响。方法:采用改良线栓法制备大鼠大脑中动脉闭塞(MCAO)再灌注模型,将Wistar大鼠随机分为正常组、假手术组、模型组、电针非经穴组和电针经穴组5组,各组随机分成ld、3d、7d 3个时间点,运用免疫组织化学技术检测下丘脑β-EP、Glu蛋白表达水平。结果:与正常组、假手术组比较,模型组各时间点指标的测定结果均有显著性差异;电针经穴可明显改善大鼠下丘脑β-EP、Glu蛋白表达水平,与电针非经穴组和模型组各时间点比较均有显著性差异。结论:电针经穴对脑缺血再灌注损伤大鼠下丘脑β-EP、Glu蛋白表达的调节具有相对特异性,电针经穴对脑缺血再灌注损伤的保护作用机制可能与其调节下丘脑β-EP、Glu蛋白表达水平有关。 Objective: To observe the influence of electroacupuncture acupoints on the protein expression of β-EP and Glu hypothalamus in rats with cerebral ischemia reperfusion injury.Methods:The model of reversible middle cerebral artery occlusion(MCAO) /reperfusion was established by improved suture method in Wistar rats.These rats were randomly divided into 5 groups: normal control group,sham-operation group,model control group,electroacupuncture acupoints group and electroacupuncture non-acupoints group.Furthermore every group was randomly divided into 3 time points 1 day,3 days and 7 days after reperfusion.The protein expression of β-EP and Glu hypothalamus in rats was observed by immunohistochemistry technique.Results: The protein expression of β-EP and Glu hypothalamus of model control group can be changed significantly compared with normal control group and sham-operation group at 1d,3d,7d of ischemia-reperfusion different points.The protein expression of β-EP and Glu hypothalamus of electroacupuncture acupoints group can be improved significantly compared with electroacupuncture non-acupoints group and model control group at ld、3d、7d of ischemia-reperfusion different points.Conclusion:Electroacupuncture acupoints regulate relative specificity of acupoints the protein expression of β-EP and Glu hypothalamus in rats with cerebral ischemia reperfusion injury.Electroacupuncture acupoints regulate the protein expression of β-EP and Glu hypothalamus which is possibly one of mechanisms of electroacupuncture acupoints protecting cerebral ischemia reperfusion injury.
