骨形态蛋白植入大白鼠皮下诱导骨形成过程中Ⅱ型胶原的分布

摘要 将骨形态蛋白(BMP)Ⅰ型胶原复合体植入到大白鼠皮下组织中,植入7、14及21日后应用免疫组织化学方法对植入物的Ⅱ型胶原的分布进行了分析。结果表明,植入7日后Ⅱ型胶原开始出现于软骨细胞周围,14日后其分布达到高峰,21日后钙化的骨基质中残留少量Ⅱ型胶原。组织学及免疫组织化学的发现表明,BMPⅠ型胶原复合体植入物所诱导的成骨过程是软骨化成骨方式。 The mixture of crude BMP and type Ⅰ collagen as a carrier was implanted in the subcutaneous tissues of 4 week old rats. The ectopic bone formation was evaluated at 7, 14 and 21 days after implantation, using HE and type Ⅱ collagen was detected within and around chondrocytes. On day 14, a large number of chondrocytes were observed with strong staining to type Ⅱ collagen, and type Ⅱ collagen, immunostaning reached its peak when evaluated both by staining intensity and staining areas. On day 21, a small amount of type Ⅱ collagen was remained in the calcified bone matrix. The results indicated that the mixture of crude BMP and type Ⅰ collagen induced ectopic bone formation through a process which it was similar to endochondral ossification.
