肝硬化形成过程中大鼠肝组织组织胺1、2受体mRNA表达的变化 被引量:1

Change in histamine-1 and-2 receptors mRNA expression during the development of experimental rat liver cirrhosis

摘要 目的研究肝硬化形成过程中大鼠肝组织组织胺1受体(H1R)和组织胺2受体(H2R)mRNA表达的变化。方法用内对照逆转录定量聚合酶链反应(RT-PCR)对经四氯化碳诱导的肝硬化形成过程中2周、4周、6周、8周(各12只)和对照组大鼠(12只)肝组织H1R和H2R的mRNA表达进行研究。结果H1R的mRNA水平:对照组(2.308±0.202)显著高于肝硬化形成过程中的2周(0.756±0.130)、4周(O.999±0.207)、6周(1.127±0.217)、8周(1.564±0.130)(P<0.01),肝硬化各组间差异有显著性(P<0.05)。H2R的mRNA水平:对照组(0.851±0.208)显著高于肝硬化形成过程中的2周(0.263±0.104)、4周(0.244±0.092)、6周(0.225±0.047)、8周(0.243±0.062)(P<0.01),肝硬化形成过程各组间差异无显著性(P>0.05)。结论大鼠肝组织以H1RmRNA的表达为主,肝硬化形成过程中肝细胞上的H1R和H2RmRNA的表达明显减少。 Objective To investigate the changes in histamine-1 receptor (H1R) and histamine-2receptor (H2R) mRNA expression in the liver tissues of the rats during the development of experimentalcirrhosis. Method Reverse transcription polymerase chain reaction (RT-PCR) was used to study theH1R and H2R mRNA expression in liver tissues in 12 control rats and 48 experimental rats (n= 12 in 2,4, 6, 8 weeks each respectively) with CC14-induced cirrhosis. Results Expression of H1R mRNA in thehepatic tissues of the control rats was 2. 308±0. 202, significantly higher than in the experimental rats(P<0. 01 ). Significant difference in H1R mRNA expression was found among the cirrhosis groups (P<0. 05). Expression of H2R mRNA in the hepatic tissues of the control rats was 0. 851±0. 208,marked1y higher than in the cirrhosis groups (P<0. 01 ). Conclusions H1R mRNA in the rat hepatictissues was mainly expressed. The expression of H1R and H2R mRNA in the hepatic tissues of the experimental rats was decreased significantly.
