摘要 选用7种树脂对磷脂酶A1(Lecitase Ultra)进行固定化,经比较得到最适的固定化载体为弱碱性大孔阴离子交换树脂D380。对固定化条件进行优化,得到最佳条件为:固定化pH 5.0,最适树脂加酶量2.0 mL/g,最适含水量7%~8%。固定化后的磷脂酶A1具有良好的热稳定性和储存稳定性,通过催化大豆磷脂与共轭亚油酸乙酯的酶促酯交换反应,确定了固定化磷脂酶A1在催化磷脂改性上的应用。 Phospholipase A1(Lecitase Ultra) was immbolized on 7 kinds of resins,and their immobilized effect were compared.Among the resins,the weak polar macroporous anion exchange resin D380 was selected for the subsequent studies.The immobilization conditions of phospholipase A1 using D380 as a carrier were optimized as follows: pH 5.0,resin adsorption capacity 2.0 mL/g and water content 7%-8%.The thermal stability and storage stability of immobilized phospholipase A1 were all improved,and could be applied in the modification of phospholipid.
