摘要 利用D-半乳糖(D-gal)致衰老小鼠模型,探讨米糠抗氧化肽对小鼠心、脑线粒体过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)、琥珀酸脱氢酶(SDH)和总三磷酸腺苷酶(T-ATPase)活性的影响,并通过聚合酶链反应(PCR)技术和光密度扫描检测该肽对小鼠脑线粒体DNA(mtDNA)缺失突变的影响。结果显示:与衰老组相比,高剂量(500mg/kgbw)米糠抗氧化肽可显著提高致衰老小鼠心脑线粒体CAT、GSH-Px、SDH以及T-ATPase活性(P<0.05),明显降低脑mtDNA缺失突变水平(P<0.01),效果优于中、低剂量。结论:米糠抗氧化肽能够提高D-gal致衰老小鼠的抗氧化能力,对mtDNA具有良好的保护作用。 The D-galactose (D-gal)-induced aging model mice were used to explore the effects of antioxidant peptides from rice bran on the activities of catalase (CAT), glutathione peroxidase (GSH-Px), succinate dehydrogenase (SDH) and total adenosine triphosphatase (T-ATPase) in mitochondria of heart and brain and on deletion mutation of mitochondria DNA (mtDNA) in brain by polymerase chain reaction (PCR) and photodensity scan. Results indicated that rice bran antioxidant peptides at the condition of high dosage could significantly increase the activities of CAT, GSH-Px, SDH and T-ATPase in mitochondria of heart and brain (P 0.05); and significantly decrease the depletion of mtDNA in brain (P 0.01) of D-gal-induced-aging mice when compared with the model group. These investigations suggest that rice bran antioxidant peptides are able to enhance antioxidant function of aging mice induced by D-gal, and have good protective effect on mtDNA.
